Buy aphrodisiac from Lee at afternoon at academy. 
Talk to Tsunade when she is cooking at day, chose 'put aphrodisiac'. Now, go to the TV room and talk to her (you can skip time in
the circle next to couch).

She can be found at:

Ichiraku ramen: Everyday at night.

For her quest, you have to accept the job offered by Teuchi (ichiraku owner). Once you accept it, you can talk to him again to skip the time till night. 
If its already night, then you can start working. 

Talk to him and Hinata will appear. Talk to Hinata for work with her (you need to talk to her twice, because first time will be a presentation).
Time will pass and you will earn 10$ and 1 point of affection with her.

For advance in her quest you need 3 point of affection. You will get her first scene and start earning +3 affection points.
For advance to the next scene you will need 12 affection points.

Keep helping Tsunade in her office, you will need 3 STEALTH point to advance scene.
You can earn stealth points by training with Anko.
You can find Anko going to the North (Where the academy is), then go to the East.

Buy sake from Rock Lee (30$).

Give it to Tsunade while she is at home in the morning (Monday and Wednesday).

When you go to sleep that day, she will visit you.


-- Spy.

You can spy at Tsunade changin in her room at morning on Tuesday and Thursday if you gave her sake the day before. (See Drunk quest)

Flower shop: everyday at afternoon

For her quest, you will need to talk to her at flower shop at afternoon and ask her for a job. 
She will ask you to bring her some rare flowers that grow in specific places. She will pay you 5$ for each one you bring.

How to get those flowers:

Go to the academy at day, go to mission room (First floor, at right)
Talk to Iruka and he will explain you some things, chose "C range" and will lend you to the first mission.

On your first mission you have to go all the way down, then right and there you can go to the second room and there is the flower you need for Ino. 
You can complete the mission or just leave, you not gonna lose the flower.
Also you can defeat some enemies for a small chance to get a flower drop and some money.

Next day, give the flower to Ino, she will ask you to bring her more.

You will need 3 flowers in total to complete her quest from 0.
